Does Hololens Companion app support API or any other interface mechanism ?
Is that possible to add some additional functionalities via API (preferably C#) to Hololens Companion app?

I do not believe the HoloLens application is extensible.However, you can write a similar application using the Windows Device Portal Wrapper. The wrapper is a C# assembly (.net and UWP) that makes it easy to call the REST APIs on developer unlocked HoloLens, Xbox One, Windows PC and Windows IoT devices.
Check out the open source Windows Device Portal Wrapper project here. The project contains a handful of samples to get started.
